Why should Edom let Yisrael pass through their land?

Rashi: Because, since Edom had not paid the debt against both of them, 1 they had no reason to begrudge Yisrael's inheriting the land, and Yisrael were therefore asking them to allow them to pass through their land without making trouble.

Why did Moshe say "Mei Be'er" (in the singular) and not 'Mei Boros'?

Rashi and Ramban (on Pasuk 19): Moshe was referring (not to Edom's water-wells, but) to the water of their own roving well, which they would refrain from using, in order to buy water - as well as food - from them. 1

Rashbam: Although it is written in the singular, Moshe was referring to the water-wells of Edom, which Moshe assured the king of Edom they would not drink, because water was scarce in that area.

Targum Onkelos: Although it is written in the singular, it means that they would not drink water from Edom's water-pits 2 .

Rashi: To the enormous benefit of Edom - as Yisrael numbered in the millions. In fact, we learn from here that a guest in a guest-house should put away his own food and purchase food from the host.

It is unclear as to why Onkelos does not translate it as 'water-wells'.

Seeing as they would go "be'Derech ha'Melech, Lo Niteh Y'min u'Semol", why did Moshe need to add "Lo Na'avor beSadeh u've'Kerem ve'Lo Nishteh Mei Be'er"?

Targum Yonasan: He was assuring them in a discreet manner that they would not seduce the virgins or rape the betrothed and married women. 1

What is the definition of "Derech ha'Melech"?

Rashbam: It means the highway that was designated for public use. 1

Seforno: It means the route which the king would designate for them. 2

Targum Yonasan: With reference to 20:17:3:1, it means that they would travel along the way of the King of the Heaven - they would behave the way that Hashem expected them to behave. 3

What did Moshe mean when he said that they would travel and not turn to the right or to the left?

1.

Rashi: He meant that they would muzzle their animals, and not allow them to eat from what grows on either side of the road.

Targum Yonasan: He meant that they would stick to the highways, and avoid traveling along the small private roads and damaging them.

Why did Yisrael request to pass through Edom, and also Sichon and Og? Seeing as the Ckoud did not go away before Aharon died, and returned immediately afterwards (refer to 25:4:4:2*), why did they not simply follow the Cloud, wherever it led them (Moshav Zekenim)?

What has one got to do through the other? Hashem would not lead Yisrael through lands belonging to others without first asking permission, and Hashem does not talk to Nochrim, so Moshe had to ask on behalf of Hashem (EC).
