Who is obligated to wash the garment?

Ba'al ha'Turim: Even though the command is issued to the Kohen, he is not obligated to actually do the washing, but others are permitted to perform it. 1

Why does the Torah write here "Vechibsu" and in Pasuk 58, "Vechubas Sheinis ... "?

Yerushalmi Shabbos, 2:1: To teach us that the first washing requires Kavanah to Tovel it ? and it will not suffice for the garment to fall into a tub of water, whereas the second washing does not. 1

What are the implications of "Eis asher bo ha'Nega"?

Rashi: It implies that also the area around the Nega 1 requires washing.

Not the entire garment, because in Pasuk 56, the Torah, inserts the word "ha'Nega" to preclude it (Sifra).
