The Kollel replies:

Tashmishei Mitzvah which are no longer fitting for their original use retain their Kedushah, so one would expect that even if the kesher were to be removed, the Retzu'os would retain their Kedushah, and henceforth there would be no difference between Retzu'os which still have a Kesher and Retzu'os which have had their Kesher removed. However, I have not been able to find a proof for this, and the possibility remains that even though the Retzu'os retain their Kedushah, they are still not on the same level as Retzu'os whose Kesher is still intact.

I looked into the question.

The only thing I have to add to the answer is derived from what the Kitzur Shulchan Aruch (10:18) says:

מעלין בקודש ולא מורידין, ותפילין של ראש קדושתה גדולה משל יד, לפי שיש בה ד'

בתים וגם השי"ן. ולכן רצועה שהיתה בשל ראש, אסור לעשותה של יד. אבל משל יד מותר

לעשותה לשל ראש. וכן רצועה של יד שנפסקה למעלה אצל הקשר, ורוצה להפכה שיעשה עתה

הקשר בקצה האחרון אסור, אלא צריך לעשות הקשר בהקצה שנפסקה שם, וכן ברצועה של

ראש, אסור להפוך מה שהיה בתוך הקשר, שיהיה חוץ לקשר. כיס שעשאו להחזיק בו

תפילין, וגם החזיקו בו, שוב אסור להשתמש בכיס זה דבר של חולין.

Based on this, I would say that there is more kedusha to the side of the strap with the knot, and therefore also more kedusha to the straps with the knot still intact, than with the knot already removed.
